Human promoter benchmark across three cell lines with bulk MPRA and scQers
Multiplex profiling of developmental cis-regulatory elements with quantitative single-cell expression reportersFive promoter constructs (noP, minP, UBCp, Pgk1p and EEF1A1p) were tested with barcode-linked reporters in HEK293T, HepG2 and K562. The table retains matched author-provided bulk RNA/DNA activity and single-cell reporter activity at the mBC/oBC level.

PiggyBac-mediated random genomic integration was used for dual RNA cassette reporters. The Pol III Tornado oBC supports reporter detection and the Pol II mBC reports promoter activity; the source table combines bulk MPRA RNA/DNA activity with scQer normalized mBC UMI activity across three human cell lines.

Quality control
The paper's high-confidence source-data rows were retained only when bulk activity and scQer activity were finite and at least 5 single-cell observations were available. Fourteen rows lacking bulk activity were excluded; the source table already applied the authors' barcode representation and reporter-detection filters.
Curation notes
The raw GSE217680 promoter count matrix is included in raw_data/geo. This processed table uses the matched source-data rows from MOESM21 so bulk and single-cell measurements remain directly aligned.
