205 full-DHS erythroid enhancer mini-library integrated lentiMPRA in HUDEP-2

Large-scale discovery of potent, compact and erythroid specific enhancers for gene therapy vectors

A pooled follow-up library of 205 full-size DHS sequences selected from the tile screen was cloned in the same GFP lentiviral screening vector and assayed in WT HUDEP-2 cells by GFP-bin sorting. The processed table contains the author-reported per-element activity scores, hg38 coordinates, and full genomic sequences supplied in Supplementary Data 4.

Processed tables are specific to each experiment. Column names, units, measurements, and table structure are not standardized across the database. Check this experiment’s column definitions and quality-control notes before comparing or combining data.

Perturbation & assay details

Untreated WT HUDEP-2 cells

This follow-up used the same integrating lentiviral reporter, minimal β-globin promoter, C1 insulator, HUDEP-2 background, and GFP-low/medium/high FACS readout as the 15K tile screen. For the mini-library, the paper estimated each full-DHS effect by linear regression of per-element sequencing counts against the mean fluorescence intensity of each GFP sorting bin. Supplementary Data 4 provides processed activity scores and sequences, but a separate per-bin count matrix for this library was not deposited in GEO.

Quality control

All 205 coordinate-unique entries supplied in Supplementary Data 4 were retained because each had a numeric activity_score and a DNA sequence whose length matched the inclusive hg38 coordinate span. No additional threshold was applied because the paper does not report a separate mini-library row-level filter.

Curation notes

Supplementary Data 4 contains 205 coordinate-unique full-DHS entries, although one Methods sentence refers to a 202-element library; the supplied data count was used. Sequence lengths match coordinate spans, including variable-length DHSs. This is a published processed-result table rather than a raw read-count matrix; the individual 40-enhancer flow-cytometry validation is retained in raw_data but is not a separate MPRA experiment.
